My latest rig uses a fender deluxe reverb amp (1976) with a Jensen P12N speaker. The guitar sound is also coming through two small cabinets (12" Jensen Neodymium speakers) powered by a Carver Cube power amp hiding behind the fender amp.
The guitar sound goes into the pedal board first (Blues driver/Ernie Ball vol/Line 6 DL4), then to a red Boss VF-1 effects half rack unit where the left side goes to the FDR and the right side goes to a Behringer MX602A mixer that is feeding the Carver stereo power amp. The effects send on the mixer goes to two TC Electronics pedals (both set on wet), first to the mono in of the Flashback/pingpong then stereo out to the stereo in of the Hall of Fame/mod and back into the stereo return of the mixer where it is mixed with the dryer guitar sound from the VF-1 right side output. The result is a big guitar sound that I can also mix as desired between the more mono sound of the fender amp and the stereo cabinets.
The guitar synth sound (left & right outputs) of the half rack Roland XV 2020 goes directly to the mixer, power amp and out to the stereo cabinets. Im using a Roland 505 synth guitar controller (24 pin cord) into a B-13 adapter (changes 24 to 13 pin) to a half rack GI-20 midi interface. I also use my Roland GM-70 interface that has the 24 pin input and makes the guitar controller more versatile. The pedal board also has a Boss FS-6 dual foot switch that controls sustain and hold functions for the guitar synth.

Here you can see the Carver Cube, mixer and TC pedals mounted in a case behind the fender amp. The lid of the case has the pedals and power transformers mounted in it and can pack up neatly when moving.
Over the years I've narrowed down all the gear I need to this set-up. It's been a
lot of trial an error but finally I can say it's all working the way I imagined it.
